exp-modificationSplenocytes were harvested and fused with P3X63 Ag8.653 murine myeloma cells using polyethylene glycol 4000. The fusion mixture was plated in 96-well tissue culture plates. The viable hybrids were screened with an ELISA. Hybridoma culture supernatants were added to coated and blocked microtiter plates containing CsCI gradient-purified virus. Positive cultures were expanded and subcloned twice by limiting dilution, and ascites fluids were produced in pristane-primed BALB/c mice.
